Estimation and Subcellular Distribution of Lecithinase Activity in Rat Intestinal Mucosa.

چکیده

A method for the quantitative estimation of lecithinase has been developed using rat intestinal mucosa as the lecithinase source. Optimal conditions for lecithinase preparation and assay involve the use of buffered glycerol media with cysteamine and EDTA to stabilize the activity. Lecithin hydrolysis proceeds with apparent first order reaction kinetics when sonicated lecithin-oleate suspensions are used : in these conditions the rate of hydrolysis is proportional to the lecithinase concentration. The method gives satisfactory results in suboellular distribution studies. Fractionation by differential centrifugation and by treatment with polymixin or pH change strongly suggest that the enzyme activity is bound to ribosomal structures.

Lecithinase and lysolecithinase of intestinal mucosa.

Enzymes, capable of hydrolysing lecithin to liberate inorganic phosphate, are present in the kidney and intestinal mucosa (King, 1931). Since several pathways may lead from lecithin to inorganic phosphate and more than a single bnzyme must have been involved, it is uncertain which enzyme was the rate-limiting one in this investigation. In a recent paper Schmidt, Bessman & Thannhauser (1957) des...

Enzymic formation of oestrone 3-glucuronide by rabbit-liver microsomes.

1. A method for the subcellular fractionation of intestinal mucosa is described. The subcellular fractions were characterized by electron microscopy and by deternining the distributions of cytochrome oxidase and alkaline phosphatase in the subcellular fractions. 2. The subcellular distribution of peptidases hydrolysing L-leucylglycine and glycylglycine has been determined in intestinal mucosa. ...

Vitamin D3 effect on the calcium content in subcellular fractions of rat intestinal mucosa.

IITOYO et al. and LAWSON et al. reported that vitamin D3 caused a slight de crease in the activities of Mg-activated DNA-dependent RNA polymerase (1, 2) and Mn++-activated DNA-dependent RNA polymerase (2). However, it is known that calcium inhibits RNA polymerase (3). Since the calcium content of the in testine in vitamin D-treated animals must be higher, the decrease in RNA poly merase activit...

Distribution and regulation of deoxythymidine kinase activity in differentiating cells of mammalian intestines.

SUMMARY The distribution of deoxythymidine kinase activity has been reexamined in rat and human intestinal mucosa separated by a planing technique into cells in different stages of differentiation. When epithelial cells leave the crypts, a decrease in kinase activity was observed in both resuspended pellet and supernatant fractions. Commercial preparations of phospholipase C added to the assay ...

Effect of an Antagonist of Vasoactive Intestinal Polypeptide on Biological Rhythm of Rest Activity in the Rat

Abstract Vasoactive Intestinal Polypeptide (VIP), has been found in different neurotransmitter systems and exists in various nerve tracts in the brain. Potential role of this peptide in physiological processes such as regulation of sleep and wakefulness, and biological rhythms has been confirmed in several reports. In the present research effects of intracerebroventricular (ICV) injection of a...
